Abstract

Pepsin significantly reduced the protein-expression efficiency of mRNA-encapsulated lipid nanoparticles (mRNA-LNPs).

  • Oral delivery of mRNA-LNPs is challenging due to instability in the gastrointestinal (GI) environment.
  • Pepsin and high concentrations of lipase were found to impair the functionality of mRNA-LNPs.
  • Pepsin treatment led to increased particle size, suggesting structural changes that affected performance.
  • LNPs formulated with C12-200, an ionizable lipid without ester bonds, showed less functional loss compared to those with SM-102 when exposed to lipase.
  • Simulated gastric fluid significantly decreased the functionality of mRNA-LNPs, primarily at pH 1 but not at pH ≥3.
