Creating Gene-Edited Pigs to Study X-Linked Immunodeficiency

Abstract

Ten IL2RG knockout male pigs were successfully generated through zygotic gene editing.

  • A mosaic mutant male piglet was obtained with a -19 bp mutation in the IL2RG gene.
  • This mutation was inherited by F1 female pigs, facilitating the development of IL2RG knockout offspring.
  • The offspring exhibited thymic tissue hypoplasia and a lack of key immune cells, indicating severe immunodeficiency.
  • The use of electroporation-mediated zygotic gene editing demonstrated feasibility for creating IL2RG-deficient pigs.
  • The ability to produce a fertile mosaic male pig capable of transmitting the mutation could offer an alternative for developing models of X-linked immunodeficiency.
