Genetic factors linked to protein changes over time in large health study groups

Abstract

In a study of 2,007 individuals, 607 proteins showed a significant genetic association with changes in protein levels over 14-18 years when baseline levels were adjusted.

  • Baseline adjustment resulted in a substantial increase in the number of proteins identified as having a genetic association with their changes over time.
  • Only 19 proteins were associated with genetic factors without baseline adjustment, with poor replication in other studies.
  • Over 70% of proteins with a genetic association for change replicated in additional datasets when baseline levels were considered.
  • Fewer than 14 proteins showed associations in longitudinal and cross-sectional models, indicating potential limitations in those approaches.
  • A majority of proteins with a genetic influence on variance also exhibited a significant change association, suggesting a link between variability and change in protein levels.
