Activating Beclin1 by Genes or Drugs May Prevent Heart and Blood Vessel Damage Caused by Aldosterone

Abstract

Aldosterone impairs endothelial function by reducing autophagic flux through mineralocorticoid receptor activation.

  • Aldosterone decreased nitric oxide production and increased reactive oxygen species in endothelial cells.
  • Impaired autophagy was indicated by elevated levels of specific autophagy markers.
  • Restoration of autophagy through Beclin1 activation improved endothelial function and normalized nitric oxide and reactive oxygen species levels.
  • Mice with enhanced Beclin1 function showed reduced cardiac fibrosis and were protected from aldosterone-induced vascular dysfunction.
  • Activation of Beclin1 via a specific peptide improved vascular function and endothelial migration impaired by aldosterone.
