Starting GLP-1 Receptor Agonists and the Risk of Sudden Pancreas Inflammation and Pancreatic Cancer: A Real-World Comparison

Essence

Starting a GLP-1RA was associated with modestly higher odds of acute pancreatitis and pancreatic cancer than starting a DPP-4 inhibitor.

Evidence

Retrospective propensity-score-matched Veterans Health Administration cohort matched 88,972 GLP-1RA initiators with 88,972 DPP-4 inhibitor initiators; acute pancreatitis was 0.31% versus 0.24% and pancreatic cancer was 0.24% versus 0.17%.

Caveat

The associations are observational and may still reflect residual or time-varying confounding despite matching and secondary analyses.
