Genetic links between GLP-1 receptor levels and idiopathic pulmonary fibrosis involving JAKMIP3 and lung function

Abstract

Higher genetically predicted GLP-1R expression is associated with a lower risk of idiopathic pulmonary fibrosis (odds ratio = 0.740).

  • The analysis showed that an increase in genetically predicted GLP-1R expression is linked to a reduced likelihood of developing idiopathic pulmonary fibrosis.
  • A positive control analysis indicated a similar association with type 2 diabetes mellitus (odds ratio = 0.630), supporting the relevance of the GLP-1R expression proxy.
  • The protein JAKMIP3 was identified as a potential mediator, accounting for 9.8% of the association between GLP-1R expression and idiopathic pulmonary fibrosis.
  • In analyses focused on pulmonary function among individuals of European ancestry, forced vital capacity showed the highest mediation proportion at 22.6%.
  • Other pulmonary function traits, including forced expiratory volume in 1 s and peak expiratory flow, also demonstrated notable mediation effects.
