How Diabetes Drugs Affect Lean Body Mass in People: A Review of Clinical Trials

Abstract

Overall, a loss of lean body mass (LBM) of 1.51 kg was observed with glucagon-like peptide-1 receptor agonists (GLP-1RA) and 1.04 kg with sodium-glucose co-transporter-2 inhibitors (SGLT2i).

  • Thirty-six studies involving individuals with obesity and other conditions were analyzed.
  • Meta-analysis indicated significant LBM loss associated with GLP-1RA and SGLT2i therapies compared to control groups.
  • Sub-group analysis showed consistent results across different body composition outcomes and measurement techniques.
  • Lean mass contributed to 28% of the total weight loss attributed to GLP-1RA and SGLT2i.
  • The findings suggest the need for monitoring body composition during weight loss treatment.
