Glutathione-Triggered Chemical Changes to Activate RNA and Extend Protein Production

Modifications to RNA can increase protein expression by up to 600% compared to unmodified RNA.

  • Disulfide-based, self-immolative acyl conjugates were designed to protect the ribose 2'-hydroxy position of RNA.
  • These modifications respond to endogenous glutathione levels, facilitating efficient RNA recovery in solution and cultured cells.
  • The shielding effect protects RNA from nuclease enzymes and translational machinery, allowing for controlled RNA release.
  • The deprotection process involves a cyclization step with varying kinetic profiles influenced by ring size and electronic properties.
  • Sustained target protein production was observed for both green-fluorescent protein and nanoluciferase in cell models over 120 hours.
