Abstract
Heparan sulfate (HS) is the most important glycosaminoglycan for lipid nanoparticle (LNP) uptake in specific cancer cells.
- The role of membrane glycosaminoglycans (GAGs) in mRNA delivery varies by nanocarrier type and cell type.
- Lipid nanoparticles require an intact glycocalyx for efficient uptake by HeLa cervical cancer cells, while cationic peptide-based formulations do not.
- HS-binding proteins, such as ApoE and vitronectin, mediate the interaction between LNPs and heparan sulfate.
- HS-dependence for LNP uptake is strong in HeLa and HCT116 tumor cells but weak or absent in primary dendritic cells and some other cell types.
- A direct correlation exists between HS-dependence and LNP uptake and expression across different cell types.
