Mice lacking E3 ubiquitin ligase RNF213 accumulate polyglucosan in the cerebellum, pons, and hippocampus.

  • Autophagy of ubiquitylated aberrant glycogen is identified as a quality control mechanism in the brain.
  • RNF213 selectively ubiquitylates abnormal glycogen to prevent its accumulation.
  • The CBM20 domain of RNF213 binds to linear oligosaccharides, limiting its activity towards normal glycogen.
  • Disruption of carbohydrate binding in RNF213 enhances its E3 ligase activity on physiological glycogen.
  • RNF213 is positioned upstream of another E3 ligase complex, LUBAC, indicating a network monitoring glycogen quality.
  • Ubiquitylated polyglucosan attracts autophagy receptors, facilitating its degradation via autophagosomes.
