Gngt2 helps inflammatory immune cells develop through an ATG5-NF-κB pathway in neutrophilic asthma

Abstract

Gngt2 expression was selectively upregulated in lung dendritic cells from mice with neutrophilic asthma.

  • Altered autophagy-related protein levels were observed in dendritic cells, including reduced ATG5 and LC3B expression.
  • In vivo silencing of Gngt2 significantly decreased neutrophilic airway inflammation and lowered levels of inflammatory cytokines IL-6, IL-23, and IL-17.
  • Gngt2 knockdown in stimulated dendritic cells restored autophagy-related markers and reduced NF-κB activation, impacting cytokine production.
  • ATG5 was identified as a downstream regulator of Gngt2, linking G protein signaling to NF-κB activation and Th17 differentiation.
  • The findings suggest that Gngt2 may contribute to immune dysregulation in type 2-low asthma through its effects on autophagy and inflammation.
