Abstract

PtdIns4P is identified as a key component of pre-autophagosomal structure (PAS) organization.

  • Pik1, a PtdIns 4-kinase, is critical for the formation of autophagosomes.
  • Localization of Atg9, Atg17, Atg1, and Atg13 to the PAS remains normal in temperature-sensitive pik1 mutant cells.
  • Recruitment of downstream Atg proteins, including components of the PtdIns 3-kinase complex I and Atg8, is impaired at restrictive temperatures.
  • PtdIns4P is transported to the PAS on Atg9 vesicles, which are derived from the Golgi.
  • Atg9 was co-immunoprecipitated with a PtdIns4P-binding protein at permissive temperatures but not at restrictive temperatures.
  • Expansion of the isolation membrane (IM) is impaired in pik1 mutant cells at the restrictive temperature.
