Changes in Gut Bacteria and Their Activity Linked to Gut Lining Problems in Healthy Family Members of Crohn's Disease Patients

Abstract

Individuals with impaired gut barrier function (lactulose-to-mannitol ratio >0.025) had reduced alpha-diversity and altered beta-diversity in their gut microbiome.

  • Reduced alpha-diversity was indicated by the Chao1 index (P = 4.0e-4).
  • Altered beta-diversity was measured using the Bray-Curtis dissimilarity index (R = 0.001, P = 1.0e-3).
  • Eight genera and 52 microbial pathways were associated with an impaired gut barrier function (q < 0.05).
  • Specifically, four genera showed decreased prevalence or increased abundance linked to impaired barrier function.
  • Eight microbial pathways related to amino acid biosynthesis were also associated with changes in gut barrier function.
  • The random forest analysis suggested a bacterial community association with gut barrier function (area under the curve, 0.63; P = 1.4e-6).
