How Gut Bacteria and Brain Support Cells May Link Depression and Type 2 Diabetes

Abstract

Gut dysbiosis is associated with both Type 2 diabetes mellitus (T2DM) and major depressive disorder (MDD).

  • Both conditions exhibit a common dysbiosis pattern characterized by lower levels of beneficial short-chain fatty acids (SCFAs)-producing bacteria and higher levels of inflammatory bacteria linked to lipopolysaccharides (LPS).
  • T2DM is associated with overactivation of astrocytes, while MDD is linked to their inhibition.
  • Microbiota dysbiosis in T2DM may trigger hyperactivation of astrocytes in the hippocampus and hypothalamus, leading to decreased glutamine synthetase and brain-derived neurotrophic factor levels, which can worsen neuroinflammation and insulin resistance.
  • In contrast, dysbiosis in MDD may suppress astrocyte activation, contributing to neuroinflammation and insulin resistance, which can exacerbate T2DM.
  • The interplay between T2DM and MDD suggests potential targets for therapeutic intervention.
