HCMV protein IE2 may cause nerve damage and hearing loss by disturbing cell growth control and metabolism

Updated

Abstract

Expression of HCMV protein IE2 led to significantly elevated auditory brainstem response thresholds in transgenic mice.

  • IE2 expression is associated with impaired auditory processing and synaptic loss in cochlear structures.
  • Transgenic mice expressing IE2 demonstrated hair cell degeneration and neuronal atrophy in auditory regions.
  • ScRNA-seq analysis revealed activation of inflammatory pathways and disruptions in mitochondrial and metabolic processes in the cochlea.
  • Transmission electron microscopy indicated severe morphological abnormalities and a reduction in mitochondrial number in spiral ganglion neurons.
  • IE2 interacts with TSC2, resulting in hyperactivation of mTOR signaling and subsequent metabolic dysregulation.
  • mTOR inhibitors significantly reduced IE2-induced auditory deficits and cellular degeneration.
