How HDAC6 activity weakens cell cleanup by disrupting microtubule stability and fusion in PBDE-47 brain toxicity

Updated

Abstract

PBDE-47 exposure led to significant neuronal loss and cognitive impairment in neonatal rats.

  • PBDE-47 was shown to increase the levels of histone deacetylase 6 (HDAC6) in a dose-dependent manner.
  • The chemical decreased the acetylation of α-tubulin, resulting in destabilized neuronal microtubules.
  • This microtubule damage impaired the transport of autophagosomes and blocked their fusion with lysosomes.
  • Neuronal injury and synaptic protein reduction were observed alongside cognitive deficits due to PBDE-47 exposure.
  • Pharmacological treatments with paclitaxel and tubastatin A restored tubulin acetylation and improved autophagic function.
  • Both treatments significantly improved learning and memory deficits in vivo, with tubastatin A showing greater potential due to its safety profile.
