Hedgehog Signaling Controls Low-Oxygen Metabolic Changes in Myeloid Leukemia Cells

Abstract

Inhibition of Smoothened (Smo) reduced cell proliferation and altered metabolic parameters in myeloid malignancy cell models under both normoxic and hypoxic conditions.

  • Smo inhibition increased PARP cleavage and decreased BNIP3 expression.
  • Downregulation of glucose transporter 1 (GLUT1) and hexokinase 2 (HK2) was observed.
  • Reduced glucose consumption, lactate production, and ATP levels were associated with Smo inhibition.
  • Changes in metabolism were linked to modulation of the AMPK-mTOR signaling pathway.
  • Smo activity may influence glycolytic remodeling in the context of hypoxia in myeloid malignancies.
