Abstract

Sustained activation of HIF-1α is associated with chronic cellular senescence and may contribute to the transition from acute kidney injury to chronic kidney disease.

  • Rapid HIF-1α expression was observed in the adaptive repair group, leading to acute senescence and subsequent recovery.
  • The maladaptive repair group exhibited delayed HIF-1α activation, which was linked to chronic senescence and progressive fibrosis.
  • HIF-1α levels were correlated with autophagy activity in both AKI repair groups.
  • In human kidney cells, silencing HIF-1α reduced senescence and fibrosis, while its overexpression had the opposite effect.
  • Analysis of renal biopsies from chronic kidney disease patients revealed elevated HIF-1α levels, which correlated with reduced kidney function and increased fibrosis.
