High-dose chemotherapy with stem cell support for children with high-risk neuroblastoma

Abstract

In a review of three randomized controlled trials involving 739 children, myeloablative therapy is associated with improved event-free survival compared to conventional chemotherapy.

  • Event-free survival showed a significant statistical difference favoring myeloablative therapy (HR 0.78, 95% CI 0.67 to 0.90).
  • Overall survival did not show a significant difference when additional follow-up data were included (HR 0.86, 95% CI 0.73 to 1.01).
  • Higher incidences of renal effects, interstitial pneumonitis, and veno-occlusive disease were observed in the myeloablative group compared to conventional therapy.
  • No significant differences were found regarding serious infections and sepsis between the treatment groups.
  • Quality of life data was not reported in the studies.
