Using Modified Hyaluronic Acid to Deliver Drugs Inside Cells Through a Cell Surface Receptor and Escape from Cellular Containers

Updated

Abstract

HA-His8 demonstrated a degree of substitution of 4.66 mol% relative to HA carboxyl groups.

  • HA-His8 showed a nanoscale hydrodynamic diameter under physiological conditions, which increased in acidic environments.
  • The buffering capacity of HA-His8 was greater than that of free His8.
  • Cellular uptake studies indicated that HA-His8 was preferentially internalized by CD44-high MDA-MB-231 cells compared to CD44-low MCF-7 cells.
  • HA-His8 exhibited reduced colocalization with acidic vesicle markers over time, suggesting decreased retention in these compartments.
  • Cytotoxicity assessments indicated that HA-His8 had low toxicity across the tested concentration range.
