The protein RBBP5 controls gene regulation that affects protein balance in cells

Updated

Abstract

Proteostasis genes exhibit ~12-hour ultradian rhythms driven by an XBP1s-dependent oscillator.

  • Transcriptional regulation of proteostasis is linked to temporal epigenome dynamics and chromatin landscapes.
  • RBBP5, an essential subunit of the COMPASS complex, is identified as a key epigenetic regulator of proteostasis.
  • Histone acetyltransferases and H3K9 acetylation do not play a significant role in the expression of dynamic proteostasis genes.
  • Loss of RBBP5 increases sensitivity to proteotoxic stress, potentially due to impaired autophagy.
  • A dynamic chromatin-associated proteomic architecture is revealed, which includes components critical for the transcriptional response to proteotoxic stress.
