Measuring cell dry mass without labels to track how aging brain immune cells absorb amyloid beta

Updated

Abstract

Dry mass measurements of microglia may serve as a potential indicator of cellular senescence and impaired phagocytosis.

  • Senescent microglia show significant molecular changes linked to decreased ability to clear amyloid-β.
  • Cellular dry mass and morphological features are identified as sensitive indicators of microglial senescence.
  • Induction of microglial senescence was achieved using optimized hydrogen peroxide treatment.
  • A strong correlation was found between dry mass and the decline in phagocytic capacity in senescent microglia.
  • Aβ-treated senescent microglia displayed lower dry mass and smaller cell size, alongside larger nuclear size compared to controls.
