Abstract

TPbeta undergoes profound agonist-induced desensitization of signalling in response to the TXA(2) mimetic U46619.

  • Agonist-induced desensitization of TPbeta involves both partial and profound mechanisms.
  • Partial desensitization is mediated by protein kinase C (PKC) phosphorylation at Ser(145).
  • Profound desensitization is associated with G protein-coupled receptor kinase (GRK)2/3 phosphorylation at Ser(239) and Ser(357).
  • Inhibition of phosphorylation at either Ser(239) or Ser(357) impairs desensitization, while mutation of both sites nearly abolishes it.
  • Desensitization affects G-protein coupling and promotes beta-arrestin association, facilitating TPbeta internalization.
