How Hu antigen R and tristetraprolin oppositely control rat bile acid transporter by affecting messenger RNA stability

Silencing HuR reduced luciferase reporter activity, while silencing TTP enhanced it.

  • The apical sodium-dependent bile acid transporter (ASBT) expression may be regulated by the stability of its mRNA.
  • HuR is associated with reduced mRNA stability of ASBT, while TTP may enhance it.
  • Increased fragments of the ASBT 3' untranslated region (UTR) shortened the mRNA half-life.
  • Binding of HuR and TTP to the ASBT 3'UTR was confirmed through gel shift assays.
  • Changes in HuR and TTP protein levels corresponded with developmental changes in ASBT expression in rats.
