Collagen III gene therapy for improving skin rejuvenation

Updated

Abstract

Essence

Human collagen III mRNA improved cellular and mouse markers of UVB-induced skin .

Evidence

This preclinical study tested hCOL3A1 mRNA in human fibroblast photoaging assays and a UVB-induced murine model measuring oxidative stress, senescence, skin barrier function, dermal thickness, collagen, transcriptomes, and toxicity or immunogenicity.

Caveat

The work did not test clinical skin rejuvenation in humans, and cell and mouse endpoints may not predict durability or safety in treated human skin.

Simplified

Key numbers

443
Increase in collagen III levels
Upregulated genes in UVB + hCOL3A group compared to UVB + vehicle group
<0.0001
Reduction in senescence markers
Significant decrease in senescence markers in mice treated with mRNA
<0.0001
Increase in dermal thickness
Significant restoration of dermal thickness after mRNA treatment
