Human brain organoid shows immune cell recruitment by microglia and immune problems linked to Alzheimer's disease

Updated

Abstract

A modular human forebrain organoid platform integrates human stem cell-derived microglia and CD8+ T cells to study Alzheimer's disease pathology.

  • Microglia are involved in clearing amyloid-beta and promoting neuron maturation.
  • Microglia also activate inflammation and recruit CD8+ T cells through specific signaling pathways.
  • This recruitment occurs via CCL4/5 signaling interacting with CCR1/5 and CXCR3 receptors.
  • Blocking CCR5 or CXCR3 can prevent T cell recruitment and influence autophagy in microglia.
  • The organoid system offers a new approach for exploring neuron-immune interactions and testing potential therapies.
