Abstract

A consistent accumulation of pathogenic Aβ species is associated with mitochondrial damage in both human and rodent models.

  • In human neural progenitor cells, early oxidative stress and changes in mitochondrial network dynamics were observed after two weeks of differentiation.
  • Significant mitochondrial dysfunction, including reduced membrane potential and increased network fragmentation, was evident after six weeks of differentiation.
  • Disruption of mitochondrial clearance was indicated by reduced localization of TOMM20 to lysosomes.
  • In 9-month-old Tg rats, elevated fission markers and mitochondrial p62 accumulation were detected, which were not present in 3-month-old Tg rats.
  • Both early and late alterations in mitochondrial homeostasis were identified in response to sustained Aβ release.
