Changing fat-related parts of switchable lipo-peptides improves cell entry and boosts mRNA delivery in lab and living models

Updated

Abstract

New xenopeptide analogues exhibit up to 85-fold higher hydrophobicity than previous carriers at neutral pH.

  • The polarity of mRNA carriers shifts significantly upon endosomal protonation.
  • Transfection efficacy is positively correlated with the number of amines in the oligoamino acids.
  • U-shaped xenopeptides demonstrate improved endosomal escape and mRNA transfection efficiency with lipophilic oligoamino acids.
  • Bundle carriers do not show enhanced transfection efficiency with additional lipophilic oligoamino acids.
  • A strong connection exists between mRNA expression and endosomal escape, as measured by calcein release.
  • In vivo studies indicate increased mRNA expression in various organs and tumor tissues after administration.
