IL-11 may worsen cisplatin kidney damage by increasing cell death through disrupted cell cleanup in kidney tubule cells

Updated

Abstract

IL-11 knockdown significantly improved renal function in a mouse model of cisplatin-induced acute kidney injury.

  • Cisplatin-induced acute kidney injury is associated with increased levels of interleukin-11, which drives tubular injury.
  • Knockdown of IL-11 reduced tubular damage and improved markers of renal function, including serum creatinine and blood urea nitrogen.
  • IL-11 activation leads to mitochondrial fragmentation and a decrease in autophagic activity due to suppressed lysosomal maturation.
  • Inhibition of the ERK1/2 pathway and overexpression of TFEB can restore lysosomal function and reduce cell apoptosis and senescence.
  • In vitro studies show that recombinant human IL-11 induces similar pathways in renal cells, linking IL-11 to kidney cell injury.
