Immune and inflammation links to cell death and heart scarring genes in heart failure with preserved pumping

Updated

Abstract

An apparent area under the curve (AUC) of 1.0000 was achieved in a murine model for a three-gene panel associated with heart failure with preserved ejection fraction (HFpEF).

  • Six candidate genes—Pparg, Ace2, Smad2, Lcn2, Ptpn11, and Sqstm1—were identified as potentially linked to fibrotic remodeling in HFpEF.
  • Gene analyses indicated connections to processes such as cellular aging and renin-angiotensin signaling.
  • In a discovery cohort of 45 samples, bootstrap correction resulted in an AUC of 0.9971, suggesting strong predictive performance.
  • Exploratory analysis revealed that whole-blood PTPN11 mRNA was lower in HFpEF patients compared to controls and was inversely associated with certain biomarkers.
  • No significant immune-cell fraction differences were found between HFpEF patients and controls after statistical corrections.
