Essence

Senolytics plus anti-PD-1 therapy showed preliminary activity in head and neck squamous cell carcinoma, and immunosenescence markers tracked poorer immunotherapy response.

Evidence

Phase 2 human trial data, single-cell immune profiling, and aged-mouse tumor models linked poorer response to immunosenescence and showed a 33.3% major pathological response rate with 4.2% grade 3-4 adverse events for senolytics plus anti-PD-1 therapy.

Caveat

The efficacy signal is preliminary because the clinical evidence comes from small phase 2 cohorts without randomized confirmation and is partly supported by mouse models.
