Stress response systems help cartilage cells adapt and survive in Metaphyseal Chondrodysplasia Type Schmid

Updated

Abstract

A 13-base pair deletion in the Col10a1 gene in mice leads to features of metaphyseal chondrodysplasia type Schmid (MCDS).

  • Hypertrophic chondrocytes (HCs) in the 13del-MCDS mice show activation of hypoxia and HIFs pathways in response to endoplasmic reticulum (ER) stress.
  • The hypoxia response is an intrinsic reaction to ER stress rather than being triggered by external factors.
  • Activation of the ATF4 protein in HCs may play a role in the response to ER stress.
  • Changes in the p53 pathway and autophagy-related protein responses coincide with alterations in chondrocyte differentiation and survival.
  • Multiple stress-response pathways are engaged to help HCs adapt to chronic ER stress.
