Interleukin 6 supports lasting T cell immune response against pancreatic cancer

Abstract

Circulating IL-6 was 100-fold higher in mice with pancreatic tumors from the KxPxCx cell line compared to those with the parental cell line.

  • Improved survival rates were observed in all instances of pancreatic tumors with elevated IL-6.
  • One cell line, KxPxCx, resulted in long-term recurrence-free survival in the majority of cases.
  • Increased levels of T cells and natural killer (NK) cells, along with decreased regulatory T cells, were detected in tumors with high IL-6.
  • The presence of elevated IL-6 led to a significant increase in lymphoid aggregates within tumors.
  • Depletion of CD4 and CD8 T cells halted tumor clearance and negated the survival advantage associated with high IL-6.
  • Delivery of IL-6 mRNA via lipid nanoparticles raised systemic IL-6 levels by approximately 50-fold, decreased tumor size, and promoted anti-tumor T cell characteristics.
