Fast shape change of fat-based nanoparticles with Omega-3 improves antioxidant protection and mitochondrial protein activity in pH-sensitive drug delivery

Updated

Abstract

A 200 ms pH-dependent structural transformation in lipid nanoparticles was observed, with a lattice parameter shrinkage from 6.14 nm to approximately 5.57 nm.

  • Lipid nanoparticles co-encapsulated antioxidants ginkgolide B and quercetin, and were functionalized for targeted neuronal delivery.
  • In vivo studies demonstrated excellent biocompatibility for nose-to-brain drug delivery in mice.
  • Multidrug-loaded lipid nanoparticles induced significant changes in gene expression related to mitochondrial function and neuroprotection.
  • In vitro experiments showed a 1.5-fold increase in ATP synthase expression, enhancing mitochondrial resilience and antioxidant enzyme function.
  • The pH-sensitive nature of omega-3 fatty acids in the nanoparticles facilitated efficient release of poorly soluble compounds.
