Three-tailed ionizable lipids appear better tuned for endosomal membrane disruption than otherwise similar two- or four-tailed designs.

Evidence

This formulation and mechanism study tested an 18-lipid bioreducible library with cytosolic delivery assays, molecular dynamics simulations, and phosphorus NMR spectroscopy.
