Isongifolene reduces intestinal cell death and improves Crohn's-like inflammation in mice

Abstract

Isolongifolene (ISO) reduced apoptosis in intestinal epithelial cells and improved multiple health indicators in a mouse model of colitis.

  • ISO alleviated body mass loss, colon length shortening, disease activity index (DAI) score, inflammatory rating, and transepithelial electrical resistance (TEER) values in mice with induced colitis.
  • ISO decreased levels of pro-inflammatory factors including TNF-α, IFN-γ, IL-1β, and IL-6 while increasing the expression of tight junction proteins ZO-1 and claudin-1.
  • In a cell model, ISO reduced the proportion of apoptotic intestinal epithelial cells and decreased the expression of apoptotic proteins cleaved-caspase-3/caspase-3 and Bax, while increasing Bcl-2 levels.
  • Activation of the AMPK/PGC1α signaling pathway was associated with the protective effects of ISO on intestinal epithelial cells.
  • The AMPK inhibitor Compound C increased apoptosis rates in ISO-treated cells and decreased levels of ZO-1 and claudin-1.
