Measuring protein production from mRNA-LNPs: how dose and previous exposure affect results

Updated

Abstract

D-luciferin delivery, imaging interval, and mouse position significantly influenced bioluminescence signal acquisition in BALB/c mice administered mRNA lipid nanoparticles.

  • mRNA dose and injection volume affected the strength and distribution of bioluminescence expression.
  • Background bioluminescence from mice receiving PBS was negligible compared to signals from mRNA lipid nanoparticles.
  • Repeated administration of lipid nanoparticles reduced expression of mRNA after subsequent doses.
  • The reduction in expression was linked to prior exposure to the lipid nanoparticle carrier, not to repeated injections or the reporter gene itself.
  • Standardization of imaging and dosing parameters is necessary for accurate bioluminescence assessments.
