Needle-free combined boosters strengthen naked mRNA vaccines to protect mice from lung viruses

Updated

Abstract

A combination of aluminum phosphate and CpG oligonucleotide substantially enhanced immune responses in a naked mRNA vaccine formulation.

  • Naked mRNA vaccines delivered through liquid jet-injection have limited immunogenicity.
  • Many tested adjuvants did not enhance humoral or cellular immunity in mouse models.
  • The aluminum phosphate and CpG oligonucleotide combination increased both humoral and cellular immune responses.
  • The adjuvanted naked mRNA vaccine induced IgG2a and cellular immunity comparable to lipid nanoparticles but with lower reactogenicity.
  • This vaccine formulation effectively eliminated infections by SARS-CoV-2 and influenza A in plaque-forming assays.
