Ketamine changes natural opioid signals and lowers heroin-seeking behavior in male rats

Updated

Abstract

Ketamine significantly reduced heroin-seeking activity in male Long-Evans rats.

  • Increased expression of genes and proteins related to the endogenous opioid system was observed.
  • Elevated levels of mu-opioid receptor proteins and enhanced G protein activity were noted, indicating increased receptor function.
  • Transcriptional changes were especially pronounced in the shell subregion of the nucleus accumbens (NAc).
  • Ketamine altered N-Methyl-D-Aspartate receptor subunit expression in both the medial prefrontal cortex and NAc.
  • The findings suggest ketamine may reduce acute relapse vulnerability to opioids through modulation of opioid and glutamatergic systems.
