Ketamine's effects on brain activity linked to repetitive negative thinking in a controlled brain imaging study

Abstract

Ketamine acutely reduced the occurrence rate of a hybrid brain network associated with rumination compared to placebo.

  • A hybrid coactivation pattern comprising the default mode and frontoparietal networks showed a positive association with rumination at baseline.
  • Higher levels of rumination were linked to larger decreases in the occurrence rate of the hybrid brain network following ketamine treatment.
  • The effects of ketamine on this brain network were transient, returning to baseline levels after 24 hours.
  • Exploratory analysis indicated that ketamine also reduced engagement of a somatomotor brain network during infusion.
  • Lamotrigine pretreatment may nominally attenuate the changes induced by ketamine, suggesting a potential glutamatergic involvement.
