Kidney problems and their treatment in people with tuberous sclerosis complex in Brazil

Abstract

Kidney complications are common in patients with tuberous sclerosis complex (TSC), particularly due to angiomyolipoma.

TSC is caused by genetic changes in the TSC1 or TSC2 genes.
The hyperactivation of the mTOR pathway is linked to the development of benign tumors in various organs.
Angiomyolipoma is the most prevalent kidney lesion in TSC patients, potentially starting in childhood.
Progression of angiomyolipoma can lead to serious issues such as hemorrhage and chronic kidney disease.
The study aims to explore the kidney health of a specific Brazilian group of TSC patients and identify risk factors for kidney-related complications.

Tuberous sclerosis complex (TSC) is a rare autosomal dominant disorder resulting from pathogenic variants in the TSC1 or TSC2 genes, leading to hyperactivation of the mechanistic target of rapamycin (mTOR) pathway and subsequent development of benign neoplasms in multiple organs. Kidney involvement is frequent, being angiomyolipoma the most common lesion. Angiomyolipoma can occur since childhood and progress throughout life, potentially leading to complications such as hemorrhage and chronic kidney disease (CKD). This study aims to characterize the kidney phenotype in a Brazilian cohort of TSC patients and identify potential risk factors associated with kidney complications.
