Detecting three special fats in horse blood as markers for long-term monitoring of gene-doping using lipid nanoparticles

Abstract

Limits of detection for three ionizable lipids in equine plasma were established at 0.05, 0.01, and 0.05 ng/mL.

  • Three ionizable lipids—ALC-0315, SM-102, and MC3—were identified as critical components of mRNA therapeutics.
  • A new liquid chromatography-high-resolution mass spectrometry method enables simultaneous detection of these lipids.
  • The detection of SM-102 in a horse's plasma lasted for up to 14 days after administration of LNP-encapsulated mRNA.
  • This method provides the longest known detection windows for LNP components in equine plasma.
  • The approach may enhance the capabilities of equine antidoping laboratories in monitoring for gene-doping.
