Reduced LC3-related clearance in immune cells from systemic sclerosis patients

Updated

Abstract

Monocyte-derived macrophages from patients with systemic sclerosis (SSc) exhibited impaired LC3-associated phagocytosis.

  • No major defects in canonical autophagy were found in lymphocytes and monocytes from SSc patients.
  • Impaired phagocytosis was assessed using IgG-coated beads triggering LC3-associated phagocytosis.
  • The proportion of LC3-positive phagosomes (LAPosomes) was lower in macrophages from SSc patients.
  • Impaired LAP in macrophages may contribute to defective clearance of dead cells, which is associated with persistent inflammation and fibrosis.
  • LAP is identified as a previously unrecognized pathway altered in SSc.
