Autophagy of misfolded procollagen in mouse bone-forming cells occurring without LC3 and GABARAP proteins

Abstract

Over 10% of type I procollagen is misfolded and rerouted from osteoblast endoplasmic reticulum to lysosomes.

  • Osteoblast-specific autophagy knockouts in mice produced only mild effects on bone formation.
  • A Gly610-to-Cys substitution in the COL1A2 chain increases misfolding of procollagen type I, leading to cell stress.
  • Disruption of autophagy did not significantly affect the degradation of misfolded procollagen in osteoblasts.
  • Live-cell imaging showed that misfolded procollagen was delivered to lysosomes via a pathway independent of certain autophagy proteins.
