The ester compound 4 demonstrated sustained conversion to its active metabolite 4a in human plasma.

  • Acid analogs of danuglipron maintained activity similar to both danuglipron and the peptide drug tirzepatide.
  • Danuglipron showed reductions in body weight and improved glycemic control but had a short duration of action and gastrointestinal side effects.
  • The study identified modifications to danuglipron that could enhance pharmacokinetic properties.
  • The potential of ester derivatives as prodrugs may lead to more sustained systemic exposure.
  • Further validation through in vivo studies is needed to confirm safety and effectiveness.
