Abstract

Dorsomedial hypothalamus neurons consist of equal populations of glutamatergic and GABAergic types.

  • Activation of glutamatergic and GABAergic dorsomedial hypothalamus neurons may increase energy expenditure and locomotion.
  • Chemogenetic activation of these neurons did not reproduce the beneficial metabolic effects seen with direct leptin receptor activation.
  • Glutamatergic leptin receptor-expressing neurons project exclusively to the raphe pallidus, while GABAergic projections overlap with different brain regions.
  • A significant proportion of leptin-inhibited neurons in the dorsomedial hypothalamus were observed, and blocking synaptic effects increased the proportion of leptin-activated neurons.
  • These findings suggest that stimulation of both GABAergic and glutamatergic neurons may mediate metabolic benefits.
