Imaging lipid nanoparticle mRNA treatments inside the body: recent progress and new possibilities

Updated

Abstract

Lipid nanoparticles (LNPs) encapsulating messenger RNA (mRNA) typically show less than 2% of internalized RNA reaching the cytosol.

  • In vivo imaging methods have been used to study LNP biodistribution and cellular uptake in various contexts.
  • LNP biodistribution and the transport of mRNA are influenced by formulation parameters and administration routes.
  • The liver and spleen are primary sites of accumulation for LNPs in systemic delivery.
  • Particles sized between 20-100 nm tend to have better cellular uptake and transport compared to larger particles.
  • Quantitative studies indicate that endosomal escape is a significant barrier to effective mRNA delivery.
  • Imaging techniques have shown that LNP organ accumulation does not directly correlate with successful protein expression.
