Using lipid nanoparticles to deliver a gene activator’s instructions for treating fatty liver disease linked to metabolism problems

Updated

Abstract

Lipid nanoparticle delivery of TFEB mRNA significantly reduced hepatic triglyceride content in mouse models of MASLD.

  • Increased hepatic TFEB expression was observed following lipid nanoparticle-mediated delivery.
  • Enhanced autophagic flux and reprogrammed metabolic pathways towards fatty acid oxidation were noted.
  • Expression of lipogenic genes was suppressed alongside the activation of genes involved in fatty acid oxidation.
  • The treatment attenuated steatosis, lowered MASLD activity scores, and decreased fibrotic remodeling.
  • TFEB may protect against lipid accumulation by activating the fibroblast growth factor 21 signaling pathway.
