Essence

Standard lipid nanoparticle ratios appear robust but not necessarily optimal for every composition-process context.

Evidence

A formulation/platform experiment compared OFAT and DoE lipid-ratio screens with manual and automated microfluidic manufacturing, finding multiple nonstandard ratios with particle size < 120 nm, PDI < 0.2, and encapsulation > 75%.

Caveat

DoE predictions were limited within the explored design space, especially for biological outputs, and manufacturing method changed formulation-performance relationships.
