Abstract

UV and oxidative stress induce pronounced chemical degradation in mRNA lipid nanoparticles (LNPs).

  • Chemical degradation in mRNA-LNPs is lipid-dependent and includes headgroup oxidation and ester hydrolysis.
  • Degradation pathways are triggered by specific environmental stressors, notably UV light and oxidation.
  • Modest heat and freeze-thaw cycling do not significantly affect the stability of these formulations.
  • Despite the degradation, particle size and encapsulation efficiency remain unchanged.
  • Understanding these degradation pathways may help in designing more stable mRNA therapeutics.
